BeWo
外观
BeWo (亦称ATCC CCL-98) 是人类胎盘绒毛膜癌细胞系[1],较常应用于胎盘屏障生理功能[2]及人类免疫缺陷病毒的研究[3],而超三倍体JEG-3细胞则是其衍生的克隆。BeWo细胞具有可产生大量层粘连蛋白的细胞外基质[4],并且BeWo细胞会表达雌激素 (雌酮、雌二醇及雌三醇)、孕激素、人绒毛膜促性腺激素、胎盘催乳素及角蛋白等物质。目前已知BeWo的b30亚克隆可在双房室 (bicameral chambers) 的可渗透膜上生长,以形成融合性的细胞层,从而确定营养从顶端表面吸收的速率,以及从基底外侧膜中流出的速率[5]。
有研究指出沉默LOX在Bewo细胞中的表达,会明显降低Bewo细胞侵袭及转移能力。因为过度表达LOX可以改变肿瘤细胞外基质的结构,并且激活特定通路及上调MMPs类的表达,使乳癌的转移侵袭速度因基底膜被降解而加速[6]。
